Danish scientists solve old blood mystery: New intriguing knowledge on blood hemoglobin

ScienceDaily (Aug. 31, 2012) ? Scientists at the research centre MEMBRANES at Aarhus University, Denmark, have completed an old puzzle, which since the 1960s from many sides has been regarded as impossible to complete. The challenge was to solve the structure of the protecting protein complex that forms when hemoglobin is released from red cells and becomes toxic. This toxic release of hemoglobin occurs in many diseases affecting red cell stability, e.g. malaria.

Technically, the most important finding in this report in Nature is a high-resolution three-dimensional mapping of the so-called 'haptoglobin-hemoglobin complex'.

"After many failing experiments, our breakthrough came when we gave up using human material and went to the local slaughterhouse to purchase pig blood. Not a particular high-technological approach, but this transition from studying human blood to blood from a species with close homology had magic effects. After running into dead ends for two years and trying out the most complex gene-technological ways to produce the right material, it suddenly worked," says S?ren Kragh Moestrup, the head of the research group at Department of Biomedicine.

The discovery provides new essential information on hemoglobin that makes up most of the red cell interior. hemoglobin is an essential blood component for transport of oxygen, but it becomes toxic with potential damaging effects on tissues, in particular the kidneys, when it is released from the red cells. An excessive release can occur in many diseases, such as malaria and other infections.

However, the body has a sophisticated defence system. The first line defence is carried out by the blood protein haptoglobin, which captures hemoglobin and gates it to a receptor that engulfs the hemoglobin-haptoglobin complex. This function of the receptor named CD163 was originally discovered by the same group.

"We have now shown how this unique protein complex forms by generation of a detailed 3-dimensional map of each atom. This shows for the first time how the complex is formed and explains the tight protein association," says PhD Christian Brix Folsted Andersen. He has together with Master's student Morten Torvund-Jensen been an essential driving force in the project.

The results have also led to an unexpected discovery of a novel type of protein structure and a new patent submission on exploitation of the discovery for use in generation of a new type of synthetic proteins to be used in therapy and diagnostics.

New York reform efforts stuck in the mud without union cooperation

NEW YORK?? The New York state Department of Education says there are 221 poorly performing schools in the state that must drastically improve within the next three years or risk being shut down.
?
More than half of those schools ? 123 ? are located in New York City.
?
The schools have until the end of September to explain to the state how they plan to improve and avoid closure. Meanwhile, New York City officials must implement a new teacher evaluation system by January or risk losing some state education funding.
?
Wait a minute ? haven?t we heard this news before?
?
Wasn?t is just last spring that New York Mayor Michael Bloomberg ?identified dozens of failing schools as ?turnaround? targets, and announced plans to replace many staff members and give students a fresh start?
?
But the United Federation of Teachers went to court to block the effort, and a friendly judge sided with them. As a result students are headed to the same old failing schools this fall.
?
Hasn?t the Bloomberg administration been trying since last year to try to design a meaningful teacher evaluation system that will purge poor instructors from the classroom and improve education for all city students?
?
Unfortunately the United Federation of Teachers has consistently refused to sign on to a new evaluation system, and Gov. Andrew Cuomo has ruled that union approval is necessary. That means the January deadline could come and go without any new system in place.
?
New York parents and taxpayers need to open their eyes. Many of their elected officials have been trying to address public education problems for several years, but the radical and self-serving UTF keeps blocking their efforts.
?
It seems the union won?t cooperate with any plan that demands more accountability from teachers. UFT leaders either believe all of their members are doing just fine, or they really don?t care how they?re doing in the classroom, as long as they pay their dues on time.
?
Real education reform will not come to New York City unless the unions are left on the sidelines and responsible adults are allowed to make the decisions.

1 dead, 2 hurt in Connecticut home explosion

An effort to fix a propane leak ends in tragedy when the house exploded, killing one man and injuring two others. WVIT's Ilana Gold reports from Connecticut.

One man?was killed?and two others were critically injured after an apparent propane explosion at a home in Connecticut Wednesday.

Police said the homeowner, John Wilkinson, 46, called his friend, Anthony Fratino, 47, to help fix a propane leak in the water heater in the basement of his home in New Milford, Conn.

The explosion happened around 6:40 p.m., according to reports.

Fratino was killed instantly, officials said. Wilkinson and Fratino's 9-year-old son, Nicholas, who was at the home at the time,?were rushed to Danbury Hospital in Danbury Conn., with severe lacerations and burns.

Nicholas, police said, will soon be transported to a hospital in Boston, more than 150 miles away.

Wilkinson is being treated at the burn unit of Bridgeport Hospital, in Bridgeport, Conn.

Wilkinson?s wife, Alice, and their two kids reportedly left the home shortly before the explosion and were at a neighbor?s house. Police investigators said Wilkinson had told his family to leave the house when he smelled propane. It was not clear where the family is staying now.

Bits of rubble were scattered over a wide area where the two-story farmhouse had stood. All that is left of the Wilkinson family home now is the chimney, NBCConnecticut.com reported.

?It?s just an incredible site; there?s nothing left of the house,? New Milford Police Department Spokesperson Lt. Lawrence Ash told The News-Times in Danbury. ?I?ve never seen anything like this.?

After the blast, police and fire crews searched the rubble for any remaining victims. While they determined no one else was at the house at the time it was leveled, Ash said police still plan to use a cadaver dog to make sure there were no human remains at the site.

Police ordered onlookers away from the scene because they feared asbestos particles from the house may be in the air, Ash said.

Some New Milford residents say they heard the explosion from miles away.

?It shook my entire house,? another neighbor, Patricia Bailey told NBCConnecticut.com. ?I had no idea what that was.?

?I thought a plane or car ran into our house,? Eric Bailey, who lives near the ruined house, told The News-Times. ?The entire house just ? boom, shook.??

Officials advise anyone who smells a gas leak in their house, workplace, or around any gas equipment or appliance to leave the area immediately and call a trained propane service person and the fire department.?

New Milford is in western Connecticut close to the New York state border.

Romney appeals to voters disillusioned with Obama

TAMPA, Florida (Reuters) - Republican Mitt Romney urged voters on Thursday to help him rebuild the U.S. economy and create millions of new jobs, asking them to overcome their disappointment in President Barack Obama and join him in restoring the promise of America.

In a high-stakes speech accepting the Republican presidential nomination, Romney said he would work to unify a divided country that believed Obama's lofty campaign promises but had lost hope they would be fulfilled.

"What is needed in our country today is not complicated or profound. It doesn't take a special government commission to tell us what America needs. What America needs is jobs. Lots of jobs," the former Massachusetts governor told a cheering crowd on the final night of the Republican National Convention in Tampa.

The speech was somewhat overshadowed by a bizarre performance on stage by Hollywood star Clint Eastwood, who addressed an imaginary Obama in an empty chair.

A surprise guest who appeared shortly before Romney during TV prime time, Eastwood made a rambling speech that puzzled viewers and interrupted a carefully crafted evening that told the story of Romney's life and why he should lead the country.

Earlier in the evening, the convention heard from a series of Romney's friends and relatives who painted a picture of a humane, compassionate man - part of a three-day effort to humanize a candidate often accused of being cold and formal.

Romney's speech, which launches a two-month dash to the November 6 election, was seen by tens of millions of television viewers and gave some their first extended look at the multimillionaire ex-businessman.

It could be a defining moment for a candidate who has struggled to win over conservatives and connect with independent voters in a campaign against Obama that has been dominated by the sluggish economy and lingering high unemployment.

Romney, who says his experience as a business executive is the cure for the ailing economy, promised to create 12 million jobs. He drew a sharp comparison between the promise of Obama's election in 2008 and the results of the last four years.

"Today the time has come for us to put the disappointments of the last four years behind us, to put aside the divisiveness and the recriminations," he said. "Now is the time to restore the promise of America."

He said Americans wanted to believe in Obama but had suffered from his failures of leadership.

"Hope and change had a powerful appeal. But tonight I'd ask a simple question: If you felt that excitement when you voted for Barack Obama, shouldn't you feel that way now that he's President Obama?"

"You know there's something wrong with the kind of job he's done as president when the best feeling you had, was the day you voted for him.

EMOTIONAL TESTIMONIALS

Democrats alternately portray Romney, 65, as a heartless corporate raider, wealthy elitist, tax evader and policy flip-flopper who should not be trusted with the keys to the White House.

To counter that image, the convention heard emotional testimonials about Romney's work as a Mormon leader that made many convention delegates in the Tampa Bay Times Forum cry.

One couple talked of how Romney befriended and comforted their dying teenage son. A woman, Pam Finlayson, recalled how he prayed with her in hospital when her premature baby daughter was close to death.

"His eyes filled with tears and he reached down tenderly and stroked her tiny back," Finlayson said.

Romney also tried to show a softer side, describing his parents and family and defending his work at Bain Capital, the private equity company that critics have accused of raiding companies and cutting jobs.

"That business we started with 10 people has now grown into a great American success story. Some of the companies we helped start are names you know," he said, naming Staples and Sports Authority.

The Obama campaign said Romney failed to lay out a path for America.

"Much like the entire Republican convention, Mitt Romney's speech tonight offered many personal attacks and gauzy platitudes, but no tangible ideas to move the country forward," Obama campaign manager Jim Messina said .

Romney's speech concluded the convention, which was cut short by a day because of a storm threat but featured stirring speeches by Romney's wife, Ann, and his vice presidential running mate, Paul Ryan.

It included a parade of new Republican faces, including several women and Hispanic speakers hoping to broaden the Republicans' appeal to voting blocs that heavily backed Obama in 2008.

It seemed to give Romney a boost. The latest Reuters/Ipsos online poll showed him moving into a narrow lead over Obama -- 44 percent to 42 percent among likely voters. The Republican had entered the week trailing Obama by four percentage points.

Obama gets his turn at the Democratic nominating convention next week in Charlotte, North Carolina, giving him an opportunity to quickly bounce back.

Romney was introduced by Florida Senator Marco Rubio, a Cuban-American with Hispanic appeal who has become a rising Republican star and a favorite of Tea Party conservatives. Rubio had the crowd roaring as he criticized Obama.

"This election is not simply a choice between a Democrat and a Republican," he said. "It's a choice about what kind of country we want America to be."

The nomination caps a political comeback for Romney, who lost the Republican race to Senator John McCain in 2008 but immediately began laying plans for a bid this year. He was tested frequently in a long and bitter primary campaign by a series of conservative alternatives from Newt Gingrich to Rick Santorum.

Despite struggling to win the hearts of conservatives, he eventually outlasted all of them, helped by huge spending on negative ads by "Super PAC" groups that fund campaign advertising to support him.

Having goals in place will make managing your finances much easier. You will have to pay it back sooner or later and the later you pay it the more you will owe. Stick to essential purchases when you?re strapped for cash, such as gas or groceries. There are smart ways to use credit cards to your advantage, but these require strict discipline.

Learn to save, live by a planned budget, balance wants and needs, set goals, live within your means, and don?t rely on credit cards. Following these guidelines will set you on a path to financial freedom. Regardless of your current situation, it?s not too late to reset the clock and start fresh.

There has been a lot of buzz lately about the FDA?s approval of two prescription medications that have been developed to help treat obesity.? There is no question that the medical community has a desire to help overweight and obese individuals in their weight loss goals.? However, there is still a huge question that looms over these approvals: Which begs the question are Belviq and Qsymia worth taking?

According to study results reported in the FDA?s announcement, participants of both studies took the medications in combination with a reduced calorie diet and exercise (hmm, wondering why they didn?t just do that without the medication).

After a year of diet, exercise, and medication (yes, that isn?t a typo ? 1 year, 12 months), Belviq users saw an average of 3.0 ? 3.7% weight loss and forty-seven of participants lost 5% of their body weight in one year.? Whereas; Qysmia which can be prescribed in two doses, utilized two separate groups for their study to compare the dosage effect.? Like the Belviq participants, Qysmia participants also were on a diet and exercise plan, and after a year, there was an average weight loss of 6.7% for the lower dose of the drug, and 8.9% for the higher dose.

While these results may seem significant at first, let?s see what these announced percentage results actually translate to using a 300 pound individual.? For example:

• If our participant is 300 pounds and took Belviq, they would lose between 11.1 pounds and 15 pounds in one year.? That?s a monthly average of .9-1.25 pounds a month (The fat is just melting away!)
• Where with Qysmia, our 300-pound participant would lose 20.1 pounds in a year on the lower dose; on the (more expensive) higher dose this participant would see a 26.7 pound weight loss.? This translates to a monthly average of 1.7-2.2 pound weight loss.? (Holy Cow Batman! We may have a winner with this one!).

I think I will have to pinch myself to get myself out of this dream/nightmare.? Based off of this information, I can take a pill AND still have to adjust my lifestyle by incorporating both diet and exercise (which I was hoping the pill would help me not have to do) and only see those results.? Oops, I forgot to mention, this drug more likely will be an out-of-the pocket expense (not covered by insurance).

Most people who are overweight or obese and weigh in at 300 pounds and start a diet and exercise plan can typically lose 1-2 pounds a week on a slow week!? So why are they getting excited about .9 to 2.2 lbs in a month by adding a costly and potentially risky pill?

Regardless of the cost associated ? either out of the pocket or via insurance ? the scariest part of these drugs are the potential side effects, such as headaches, dizziness, nausea, fatigue, dry mouth, constipation, an increase in heart rate, tingling of the hands and feet, insomnia, and altered taste sensation.? Additionally it was noted: women of child-bearing age who would be prescribed Qysmia also?need to take a pregnancy test before starting the medication and every month while they are on it, and should use contraception to avoid pregnancy complications.

Credit Agricole close to selling off Greek unit

PARIS (AP) ? French bank Credit Agricole says it is close to reaching a deal to sell its troubled Greek subsidiary Emporiki.

BUENOS AIRES, Argentina (AP) ? Saving the whales is something Argentines take so seriously that authorities are planning to shoot seagulls that have developed a habit of attacking the huge marine mammals.

Environmentalists say the plan is misguided. They say humans are the real problem, creating so much garbage that the gull population has exploded, endangering the whales.

Both sides agree that what was bizarre animal behavior a decade ago has now become a real hazard for threatened southern right whales in one of their prime birthing grounds, turning whale-watching from a magical experience to something from a horror movie.

Seagulls off the coast of the Patagonian city of Puerto Madryn have discovered that by pecking at the whales as they come up for air, they can create open wounds. Then, each time the whales surface, it's dinner time: Gulls swoop down and dig in, cutting away skin and blubber with their beaks and claws.

"It's not just that the gulls are attacking the whales, but that they're feeding from them, and this way of feeding is a habit that is growing and becoming more frequent," said Marcelo Bertellotti, who works for the National Patagonia Center, a government-sponsored conservation agency. "It really worries us because the damage they're doing to the whales is multiplying, especially to infant whales that are born in these waters."

Whales also are changing their behavior in response: Instead of breaching the water and dramatically displaying their tails, they rise just barely enough to breathe through their blow-holes before descending to safety, Bertellotti said.

Bertellotti's answer: Shoot the gulls that display this behavior with air rifles and hunting guns, and recover each downed bird before they are eaten along with the ammunition, causing still more damage to marine life. His "100-day Whale-Gull Action Plan" was approved by the government of Chubut, and provincial officials came out Tuesday in defense of it.

"We are preparing a pilot plan that seeks to stop the damage from the gulls that pick at the flesh of the whales, because this is putting at risk the resource. It will be a minimal intervention to protect the life of the southern right whale and thus provide a response to the complaints of the sightseeing businesses that operate in the place," Gov. Martin Buzzi posted on his Facebook page.

Whale-watching is big business for Chubut. Southern right whales have recovered to about 8 percent of their original population since becoming a protected species worldwide, and hundreds come to the relatively calm and warm waters of the gulf formed by the Valdez Peninsula to give birth and raise their newborns each July to December.

Seeing them surface from nearby boats can be a magical experience, and gull attacks were rare until about eight years ago, said Milko Schvartzman, who coordinates the oceans campaign for Greenpeace in Latin America. But more gulls have caught on, and the population has boomed to the point where whales are attacked at least every fourth time they surface, he said.

Now the tourists are suffering along with the whales, Schvartzman said. "It's not so pleasant anymore."

Environmentalists say the only way to effectively reduce the seagull population is to deny the birds food by closing open-air garbage dumps around the gulf and stopping fishermen and a nearby seafood packing plant from dumping scraps into the water. Activists have been lobbying Chubut for many years to develop plans to reduce, recycle and properly contain garbage, but politicians have resisted, Schvartzman said.

Chubut's environmental minister, Eduardo Maza, blamed the problem on previous governments, and said the province is now working on permanent solutions. Shooting the gulls "is surely not the most pleasant measure, but it's necessary to do something to control a situation that has been growing after many years of inaction," Maza said.

"At year's end, we're going to inaugurate garbage-separation plants," Maza said. "All the garbage in the protected Peninsula Valdes area that isn't recyclable will be properly disposed of, which will enable us to mitigate the open-air garbage dumps."

Schvartzman said that if humans don't solve the problem quickly, the whales will simply stop coming.

BOGOTA, Colombia (AP) ? President Juan Manuel Santos said Monday that his government has opened "exploratory talks" with Colombia's main rebel groups in a bid to end a stubborn half-century-old conflict.

Santos also mentioned that Colombia's No. 2 rebel band, the National Liberation Army known by its Spanish-language initials ELN had expressed a desire to participate in peace talks.

The ELN has also held failed peace talks with the government in the past.

Researchers say current interventional therapies for autism among adolescents and young adults are not supported, or refuted, by existing outcome evidence.

Vanderbilt University scientists say the dearth of evidence calls for an immediate round of structured interventions that can be accurately tracked for success or failure.

Although the prevalence of autism is on the rise, much remains to be discovered when it comes to interventions for this population, the researchers concluded.

?Overall, there is very little evidence in all areas of care for adolescents and young adults with autism, and it is urgent that more rigorous studies be developed and conducted,? said Melissa McPheeters, Ph.D., M.P.H., and senior author of the report.

The report is a systematic review of therapies published by the Department of Health and Human Services? Agency for Healthcare Research and Quality (AHRQ).

Zachary Warren, Ph.D., director of the Vanderbilt Kennedy Center?s Treatment and Research Institute for Autism Spectrum Disorders, said, ?There are growing numbers of adolescents and adults with autism in need of substantial support.

?Without a stronger evidence base, it is very hard to know which interventions will yield the most meaningful outcomes for individuals with autism and their families.?

As a basis for the report, researchers systematically screened more than 4,500 studies and reviewed the 32 studies published from January 1980 to December 2011 on therapies for people ages 13 to 30 with autism spectrum disorders.

They focused on the outcomes, including harms and adverse effects, of interventions, including medical, behavioral, educational and vocational.

• Some evidence revealed that treatments could improve social skills and educational outcomes such as vocabulary or reading, but the studies were generally small and had limited follow-up;
• Limited evidence supports the use of medical interventions in adolescents and young adults with autism. The most consistent findings were identified for the effects of antipsychotic medications on reducing problem behaviors that tend to occur with autism, such as irritability and aggression. Harms associated with medications included sedation and weight gain;
• Only five articles tested vocational interventions, all of which suggested that certain vocational interventions may be effective for certain individuals, but each study had significant flaws that limited the researchers? confidence in their conclusions.

The researchers? findings on vocational interventions will be featured in the Aug. 27 issue of Pediatrics.

Magical, Fulfilling Relationships, Part II - Have a Magnificent Day!

Do you have the kind of relationships you want?
When you are with someone you are attracted to are you present in the moment with them?
Do you fully associate to the joy and fulfillment available?

I know it seems like a simple question, but most people are so busy thinking about things while they are with someone else.? Like, what am I going to say, do they like me, when should I make my move?? Does this sound familiar? Maybe you have your own questions or chatter.? I am amazed at how many people are texting and playing with their cell phones while with another person.

My question to you is, if you are busy thinking about anything when you are with the person you are with and maybe even attracted to, how can you be fully present with them?? Because if you are thinking, or concerned about how it?s going, then you are not really engaged in your energy to being with the other person.? Magic happens in those special, little moments that most people miss because they are busy thinking or worried about how they are doing.

That?s why this picture is a great reminder?are you in the past, present or future when you are asking someone for a date or with them spending ?quality time.?? Maybe your thinking about something you have to do later.? Maybe something from earlier the day is nagging at you.? Remember, the point of greatest power is always the present moment.

So how important is it for you to be present to experience true communication, clarity and connect with another person?? What do you think the other person is feeling when they sense you are not really listening to them or are not connecting with them?? Maybe they think you do not really like them?? How often do you think these factors affect the quality of your relationships and outcomes?

By the way, these patterns do not just affect romantic relationships.? Look at how they affect family interactions between parents and their children.? How about between relatives??
